Actilosa Capsules 10s is a fast-acting, effective antimotility anti-diarrheal formulation containing Loperamide HCl 2mg to quickly slow intestinal transit, reduce loose stool frequency, and restore normal digestive rhythm.

Overview & Mechanism of Action

  • Actilosa Capsules (10s pack) contains Loperamide Hydrochloride 2mg, a synthetic opioid derivative designed specifically for localized action on the gastrointestinal tract without significant central nervous system effects. It acts through targeted intestinal pathways:
  • Antimotility Action ($\mu$-Opioid Receptor Binding): Binds to $\mu$-opioid receptors located in the myenteric plexus of the intestinal wall. This inhibits the presynaptic release of acetylcholine and prostaglandins, decreasing smooth muscle tone and slowing intestinal peristaltic contractions.
  • Enhanced Fluid & Electrolyte Absorption: By prolonging gastrointestinal transit time, it allows the mucosal lining to reabsorb water, sodium, and essential electrolytes from the intestinal lumen, significantly reducing fluid volume and loose stool consistency.
  • Anal Sphincter Tone Support: Increases resting tone of the anal sphincter, helping reduce fecal urgency and incontinence during acute episodes of bowel distress.

Clinical Indications

  • Actilosa Capsules 10s is indicated for:
  • Fast symptomatic relief of acute non-specific diarrhea in adults and adolescents
  • Control and management of traveler's diarrhea caused by dietary changes or pathogens
  • Symptomatic control of chronic diarrhea associated with inflammatory bowel conditions under physician supervision
  • Reducing the frequency and volume of discharge in patients with ileostomies

Usage Guidance & Safety Precautions

  • Administration: Swallow capsules whole with water. Rehydration is critical during diarrheal illness; take alongside Oral Rehydration Salts (ORS) or plenty of fluids

Recommended Dosage:

  • Adults & Children over 12 years: Initial dose of 2 capsules (4mg), followed by 1 capsule (2mg) after each subsequent loose stool. Do not exceed 8 capsules (16mg) within a 24-hour period.Children under 12 years: Not recommended unless under direct medical prescription.

Precautions & Warnings:

  • Discontinue use if clinical improvement is not observed within 48 hours, or if constipation, abdominal distension, or ileus develops. Contraindicated in conditions where slowing intestinal motility must be avoided, such as acute dysentery characterized by blood in stool and high fever, pseudomembranous colitis, or active ulcerative colitis flare-ups.
