Adol 125mg Suppositories 10s is a fast-acting infant antipyretic and analgesic containing Paracetamol (125mg), formulated for gentle rectal administration in infants and toddlers to reduce fever and relieve pain when oral administration is impractical.

1

Overview & Mechanism of Action

  • Adol 125mg Suppositories (10-suppository pack) provides reliable fever reduction and pain management for infants and young children through direct rectal absorption:
  • Central Prostaglandin Inhibition (Paracetamol 125mg): Acts by inhibiting cyclooxygenase ($\text{COX-1}$ and $\text{COX-2}$) enzymes in the central nervous system, suppressing central prostaglandin ($\text{PGE}_2$) synthesis to raise the pain threshold and lower the elevated hypothalamic thermoregulatory set point.
  • Optimal Rectal Route for Infants: Bypasses the upper gastrointestinal tract, providing a reliable alternative for infants unable to retain oral liquid syrups due to acute vomiting, gastroenteritis, reflux, or fussiness.
  • Targeted Infant Relief: Rapidly alleviates post-immunization pyrexia, fever from viral or upper respiratory tract infections, and mild discomfort associated with infant teething and earache.

Clinical Indications

  • Adol 125mg Suppositories 10s is indicated for:
  • Rapid reduction of fever and post-vaccination febrile reactions in infants and toddlers
  • Relief of mild-to-moderate pain associated with teething, earaches, sore throat, and post-procedural discomfort
  • Alternative antipyretic and analgesic therapy for infants unable to swallow or retain oral medications due to nausea, vomiting, or acute gastrointestinal distress

Usage Guidance & Safety Precautions

Administration:

  • Thoroughly wash hands before and after application. Uncap the individual foil wrapper and remove the suppository. Moisten the tip with water or a water-soluble lubricant. Gently insert the suppository tip-first into the infant's rectum while the infant lies on their back or side with knees raised towards the chest. Gently hold the buttocks closed for a few moments to prevent expulsion.

Recommended Dosage:

  • Infants and Toddlers 3 months to 1 year (approx. 5 to 10 kg): 1 suppository (125mg) every 4 to 6 hours as needed. Do not exceed 4 suppositories (500mg total) within a 24-hour period.
  • Weight-Based Dosing: Standard pediatric dosing is $10\text{--}15\,\text{mg/kg}$ per dose every 4 to 6 hours, up to a maximum daily dose of $60\,\text{mg/kg/day}$.

Precautions & Warnings:

  • For rectal administration only. Do not administer orally. Do not combine with other paracetamol-containing products (such as oral syrups, drops, or combination cold medicines) to avoid accidental overdose and risk of severe liver damage. Contraindicated in patients with severe hepatic impairment, severe renal failure, or active rectal inflammation/proctitis. Consult a physician if fever lasts longer than 3 days or pain persists for more than 2 days. Store below $25^\circ\text{C}$ in a cool, dry place away from direct heat to prevent melting. Keep out of reach of children.
