  • Central Prostaglandin Inhibition (Paracetamol 250mg): Acts primarily by inhibiting cyclooxygenase ($\text{COX-1}$ and $\text{COX-2}$) enzymes in the central nervous system, reducing central prostaglandin ($\text{PGE}_2$) synthesis to elevate pain threshold and lower the hypothalamic heat-regulating set point.
  • Rectal Absorption Alternative: Bypasses the upper gastrointestinal tract, making it an optimal therapeutic alternative for children experiencing severe nausea, acute gastroenteritis, persistent vomiting, or post-operative oral fluid restriction.

Usage Guidance & Safety Precautions

Administration:

  • Wash hands thoroughly before and after use. Remove the suppository from its foil wrapping. Moisten the tip with a drop of water or water-soluble lubricant. Gently insert the suppository tip-first into the child's rectum while the child lies on their side with knees bent. Gently hold the buttocks together for a few seconds to ensure retention.

Recommended Dosage:

  • Children 1 to 5 years (approx. 10 to 20 kg): 1 suppository (250mg) every 4 to 6 hours as needed. Do not exceed 4 suppositories (1000mg total) within a 24-hour period.
  • Dosing by Body Weight: Standard pediatric dosing is $10\text{--}15\,\text{mg/kg}$ per dose every 4 to 6 hours up to a maximum of $60\,\text{mg/kg/day}$.

Precautions & Warnings:

  • For rectal administration only. Do not administer orally. Do not combine with other paracetamol-containing products (oral syrups, drops, or tablets) to avoid accidental overdose and risk of severe hepatic injury. Contraindicated in severe hepatic impairment, severe renal impairment, or active proctitis/rectal inflammation. Consult a doctor if fever persists for more than 3 days or pain lasts for more than 2 days. Store below $25^\circ\text{C}$ in a cool, dry place. Protect from direct heat to prevent melting.
