Overview & Mechanism of Action

• Clarityne 10mg Tablets (strip of 10 tablets) provides continuous 24-hour non-drowsy protection against indoor and outdoor environmental allergens:
• Selective Peripheral $H_1$-Receptor Antagonism (Loratadine): Functions as a long-acting tricyclic second-generation antihistamine that selectively blocks peripheral $H_1$ histamine receptors. By preventing circulating histamine from binding to vascular endothelial and smooth muscle receptors, it halts early and late allergic responses.
• Suppression of Inflammatory Cascade: Inhibits the release of histamine and pro-inflammatory cytokines from mast cells and basophils, effectively suppressing nasal congestion, ocular tearing, persistent sneezing, and histamine-induced cutaneous wheals.
• Non-Sedating Central Profile: Demonstrates minimal blood-brain barrier penetration and low affinity for central nervous system histamine receptors, providing potent peripheral allergy relief without causing daytime sedation or cognitive impairment.

Clinical Indications

• Clarityne 10mg Tablets 10s is indicated for:
• Rapid, 24-hour relief of symptoms associated with seasonal allergic rhinitis (hay fever) and perennial allergic rhinitis, including sneezing, rhinorrhea, nasal pruritus, and ocular itching
• Treatment of chronic idiopathic urticaria (hives), reducing skin itching, red wheals, and localized cutaneous swelling
• Relief of allergic skin conditions accompanied by pruritus and inflammation

Usage Guidance & Safety Precautions

Administration:

• Swallow the tablet whole with a glass of water. May be taken with or without food.

Recommended Dosage:

• Adults & Children over 12 years (or weighing over 30 kg): 1 tablet (10mg) once daily.
• Children 2 to 12 years (weighing under 30 kg): Pediatric syrup or lower-dose formulations are recommended.

Precautions & Warnings:

• Generally non-drowsy; however, individual sensitivity varies, so exercise caution when driving or operating machinery until personal response is established. Dose reduction (e.g., 10mg every other day) is recommended in patients with severe liver impairment. Store below 30°C in a dry place protected from direct heat and moisture. Keep out of reach of children.
